CBD Reduced Isolation-Related Aggression in Mice, but Human Evidence Is Still Limited

A 2019 study found that cannabidiol (CBD) reduced aggressive behavior triggered by social isolation in male mice. The findings suggest a possible biological pathway for CBD’s effects, but they do not show that CBD can safely or effectively treat aggression in people.

In the study published in Progress in Neuro-Psychopharmacology and Biological Psychiatry, researchers housed male Swiss mice individually for 10 days. This isolation protocol was used to increase aggression. The animals were then tested using a “resident-intruder” procedure, in which an unfamiliar mouse was placed in the resident mouse’s cage.

The researchers administered CBD at doses of 5, 15, 30 or 60 milligrams per kilogram before the behavioral test. All four doses reduced the number of attacks compared with the control condition. The 15- and 30-mg/kg doses also increased the time before the first attack and shortened the duration of aggressive encounters. None of the tested doses appeared to impair the mice’s general locomotor activity.

Additional experiments suggested that the effect involved both the serotonin 5-HT1A receptor and the cannabinoid CB1 receptor. When researchers administered drugs that blocked either receptor, CBD’s anti-aggressive effect was weakened. The study also reported reduced activity of a neuronal marker in the lateral periaqueductal gray, a brain region involved in defensive and social behaviors.

These results do not support the original suggestion that higher CBD doses increased aggression. Within the dose range tested, CBD reduced the number of attacks at every dose, although the effects on attack latency and encounter duration were strongest at intermediate doses. That pattern highlights the importance of dose selection without proving that a particular dose would work in humans.

The study was conducted entirely in mice using aggression induced by a specific period of social isolation. Animal behavior models can help researchers investigate potential mechanisms, but they cannot establish that CBD will treat aggression associated with autism, psychosis, personality disorders or other psychiatric conditions. Human trials would be needed to assess effectiveness, appropriate dosing and possible effects on different types of aggression.

CBD also is not risk-free. The U.S. Food and Drug Administration says that the only FDA-approved CBD drug, Epidiolex, is indicated for certain seizure disorders—not aggression. Clinical information summarized by the National Center for Complementary and Integrative Health lists possible side effects including decreased appetite, diarrhea, sleepiness, drug interactions and liver-related risks. Over-the-counter CBD products may also vary in strength and quality.

For now, the mouse study is best viewed as preliminary evidence that CBD can influence isolation-related aggression in an animal model. It offers a basis for further research, not a clinical recommendation to use CBD as a treatment for aggressive behavior.
