CBD Improved Memory and Reduced Neuroinflammation in an Alzheimer’s Mouse Model

Could cannabidiol (CBD) help protect the brain from processes linked to Alzheimer’s disease? A 2023 study in Cells found that CBD reduced cognitive impairment and signs of neuroinflammation in mice exposed to amyloid-beta, but the findings remain preliminary and do not show that CBD prevents or treats Alzheimer’s disease in people.

The study by researchers at China Pharmaceutical University, published on November 21, 2023, examined CBD in cell experiments and in a mouse model created by administering aggregated Aβ1–42, a form of amyloid-beta associated with Alzheimer’s research. The researchers also used RNA sequencing to investigate changes in gene activity after CBD treatment.

In cultured SH-SY5Y nerve-like cells, CBD at the concentrations tested did not produce notable toxicity and partly protected the cells from damage caused by Aβ1–42. In the mouse experiments, animals exposed to amyloid-beta performed worse on the Morris water maze, a test of learning and spatial memory. Mice treated with CBD showed shorter escape times, suggesting an improvement in the memory deficit produced by the experimental treatment.

The researchers also observed changes in several markers of brain inflammation. CBD reduced levels of inflammatory molecules including tumor necrosis factor alpha and monocyte chemoattractant protein-1. It also reduced activation of microglia and astrocytes in parts of the hippocampus and cortex. These cells normally support and protect the brain, but excessive or prolonged activation can contribute to inflammatory damage.

RNA sequencing pointed to broader changes in genes and biological pathways involved in inflammatory responses. The findings are consistent with the idea that CBD may influence Alzheimer’s-related inflammation and support neurotrophic signaling, rather than acting only through the amyloid pathway.

However, the study has important limitations. It used laboratory cells and mice exposed to injected amyloid-beta, not people with Alzheimer’s disease or a long-term genetic model that reproduces every feature of the condition. Improvements on a mouse memory test also cannot be assumed to translate into improved cognition in humans. The study’s CBD dose—25 milligrams per kilogram in mice—does not establish a safe or effective human dose, and animal doses cannot be converted directly into self-treatment instructions.

CBD also has established safety considerations. The U.S. Food and Drug Administration notes that CBD can cause liver injury, interact with medications and produce sedation. Apart from the prescription CBD drug approved for certain rare seizure disorders, CBD products have not been evaluated by the FDA as treatments for Alzheimer’s disease.

There is currently no evidence that over-the-counter CBD products prevent dementia or slow Alzheimer’s disease in humans. The Alzheimer’s Drug Discovery Foundation describes the existing evidence as predominantly preclinical and notes that larger, longer clinical trials are needed to assess CBD’s effects on cognition.

Alzheimer’s treatment has also advanced since many early CBD reports were published. According to the National Institute on Aging, lecanemab and donanemab are FDA-approved for selected people with early Alzheimer’s disease, while other medications can help manage symptoms. These treatments do not cure the disease, but they illustrate why potential therapies must be evaluated in carefully designed human trials.

The China Pharmaceutical University study provides useful evidence that CBD can affect amyloid-related toxicity, inflammatory signaling and memory behavior in experimental models. It supports further research into CBD and related compounds, but it is not a clinical demonstration of Alzheimer’s prevention or treatment. Until human studies establish its benefits, dosage and risks, people with Alzheimer’s should not replace prescribed care with CBD without speaking with a qualified health professional.
