Hemp Hull Compounds Show Promise for Gut Barrier Support in Cell-Based Research
Two phenolic compounds found in hemp seed hulls may help protect intestinal barrier function, according to a 2022 laboratory study. The findings are promising but preliminary: the research was conducted in cultured human colon cells, not in people.
In the study published in the Journal of Food Bioactives, researchers examined N-trans-caffeoyltyramine (NCT) and N-trans-feruloyltyramine (NFT), naturally occurring compounds identified in hemp hulls. The work was conducted by researchers from Brightseed, the biosciences company that developed the ingredient examined in the research.
The intestinal barrier helps regulate the movement of nutrients into the body while limiting the passage of harmful substances, including toxins and pathogens. Inflammation can weaken this barrier and increase intestinal permeability. Although “leaky gut” is widely used in consumer health discussions, intestinal permeability is a complex biological measure and should not be treated as a diagnosis on its own.
To model inflammation, the researchers exposed cultured human transverse colon epithelial cells to tumor necrosis factor, or TNF. They then treated the cells with NCT, NFT, or a combination of the two for 48 hours. TNF reduced transepithelial electrical resistance (TEER)—a laboratory indicator of barrier integrity—and increased permeability. The combined NCT and NFT treatment produced a dose-dependent improvement in both measures, with statistically significant effects under the conditions tested.
These results suggest that NCT and NFT may influence intestinal barrier function during an inflammatory challenge. However, the experiment did not show that eating hemp hulls, taking a supplement, or using a hemp-derived fiber product prevents or treats a digestive disorder. Cell-culture findings cannot establish effectiveness, appropriate dosage, safety for particular medical conditions, or how the compounds are absorbed and metabolized in humans.
Hemp hulls also contain substantial dietary fiber and other nutrients. Follow-up laboratory research published in 2024 found that a hemp-hull fiber ingredient and purified NCT and NFT altered the composition and activity of human gut microbial communities in vitro, including changes in microbial diversity and short-chain fatty-acid production. That study likewise did not demonstrate a clinical benefit in humans; it provides additional mechanistic evidence for further investigation.
Some human research has since examined a hemp-hull fiber product for digestive comfort. In a 2024 randomized, placebo-controlled trial involving healthy adults with digestive discomfort, participants receiving the product reported better outcomes on several measures than those receiving inulin at selected time points. The trial evaluated symptoms and quality of life—not whether NCT and NFT repaired an impaired intestinal barrier—and its findings should not be generalized to the treatment of gastrointestinal disease.
Overall, the evidence supports describing hemp hulls and their bioactive compounds as an area of research rather than as a proven digestive-health therapy. Larger, independently replicated human studies are needed to determine whether NCT, NFT, or hemp-hull fiber can meaningfully improve gut barrier function or digestive symptoms in specific populations.
