Raphael Mechoulam’s Lasting Legacy in Cannabis and Endocannabinoid Research

Raphael Mechoulam, the Israeli chemist widely regarded as a founder of modern cannabinoid research, died on March 9, 2023, at age 92. His work transformed cannabis from a poorly understood plant into a subject of rigorous chemical, pharmacological, and medical research.

Born in Sofia, Bulgaria, in 1930, Mechoulam immigrated to Israel in 1949. He studied biochemistry at the Hebrew University of Jerusalem, earned his doctorate at the Weizmann Institute of Science, and later joined the Hebrew University faculty. He began investigating cannabis chemistry in the early 1960s, first at the Weizmann Institute and later at the Hebrew University’s School of Pharmacy.

One of his most important early achievements came in collaboration with Yechiel Gaoni. In 1964, they published their work on the isolation, structure, and partial synthesis of THC, the principal intoxicating compound in cannabis. The discovery helped explain which molecule was responsible for much of cannabis’s psychoactive effect and provided researchers with a defined substance they could study in controlled experiments.

Mechoulam’s research also clarified the chemistry of cannabidiol, or CBD. Although CBD had been isolated before Mechoulam’s work, he and Youval Shvo reported the structure of cannabidiol in 1963. CBD does not produce the intoxicating effects associated with THC, but its pharmacological properties later became the subject of extensive research.

His most far-reaching contribution came decades later. In 1992, Mechoulam and colleagues identified anandamide, the first widely recognized endogenous cannabinoid—a signaling molecule produced naturally in the body. Their study in Science showed that anandamide binds to cannabinoid receptors, helping establish the foundation for what became known as the endocannabinoid system. The system is now understood to play a role in numerous physiological processes, although many of its functions and possible medical applications remain under investigation.

Mechoulam also helped advance the development of synthetic cannabinoids. Dronabinol, a synthetic form of delta-9-THC, is used in approved medicines for chemotherapy-related nausea and vomiting and for anorexia associated with weight loss in people with AIDS. The U.S. Food and Drug Administration’s overview of cannabis-related medicines distinguishes these approved indications from the many unproven claims surrounding commercial cannabis products.

Over a career spanning more than six decades, Mechoulam published hundreds of scientific papers, mentored generations of researchers, and received major honors including the Israel Prize, the Harvey Prize, and the National Academy of Sciences Award in Chemical Sciences. The Hebrew University’s tribute described his influence on cannabinoid science as extending far beyond any single discovery.

Mechoulam’s legacy is not simply that he identified important molecules. He helped create the scientific framework that made it possible to study how cannabinoids act in the plant, in medicines, and within the human body. His discoveries continue to shape research into pain, epilepsy, inflammation, neurological disorders, and other areas—while also reminding us that promising findings require careful clinical study before they become established treatments.
